    Immobilization of Shewanella oneidensis MR-1 in diffusive gradients in thin films for determining metal bioavailability

    Assessing metal bioavailability in soil is important in modelling the effects of metal toxicity on the surrounding ecosystem. Current methods based on diffusive gradient thin films (DGTs) and Gel-Integrated Microelectrode are limited in their availability and sensitivity. To address this, S. oneidensis, an anaerobic iron reducing bacterium, was incorporated into a thin layer of agarose to replace the polyacrylamide gel that is normally present in DGT to form biologically mobilizing DGT (BMDGT). Viability analysis revealed that 16-35% of the cells remained viable within the BMDGTs depending on the culturing conditions over a 20 h period with/without metals. Deployment of BMDGTs in standardized metal solutions showed significant differences to cell free BMDGTs when cells grown in Luria Broth (LB) were incorporated into BMDGTs and deployed under anaerobic conditions. Deployment of these BMDGTs in hematite revealed no significant differences between BMDGTs and BMDGTs containing heat killed cells. Whether heat killed cells retain the ability to affect bioavailability is uncertain. This is the first study to investigate how a microorganism that was incorporated into a DGT device such as the metal reducing bacteria, S. oneidensis, may affect the mobility of metals

    Honeybee nutrition is linked to landscape composition

    Declines in insect pollinators in Europe have been linked to changes in land use. Pollinator nutrition is dependent on floral resources (i.e., nectar and pollen), which are linked to landscape composition. Here, we present a stratified analysis of the nutritional composition of beebread in managed honeybee hives with a view to examining potential sources of variation in its nutritional composition. Specifically, we tested the hypothesis that beebread composition correlates with local land use and therefore available floral resources. The results demonstrated that the starch, lipid, and moisture contents of beebread are all highly conserved across hives, whereas levels of protein and nonreducing sugar increased as the year progressed, reducing sugars, however, decreased during the first half of the year and then increased toward the end. Local land use around hives was quantified using data from the Countryside Survey 2007 Land Cover Map. Bee-bread protein content was negatively correlated with increasing levels of arable and horticultural farmland surrounding hives and positively correlated with the cover of natural grasslands and broadleaf woodlands. Reducing sugar content was also positively correlated with the amount of broad-leaved woodland in a 3Km(2) radius from the hives. Previous studies on a range of invertebrates, including honeybees, indicate that dietary protein intake may have a major impact on correlates of fitness, including longevity and immune function. The finding that beebread protein content correlates with land use suggests that landscape composition may impact on insect pollinator well-being and provides a link between landscape and the nutritional ecology of socially foraging insects in a way not previously considered

    Mycobacterium avium subspecies paratuberculosis:human exposure through environmental and domestic aerosols

    Mycobacterium avium subspecies paratuberculosis (Map) causes Johne’s disease in animals and is significantly associated with Crohn’s disease (CD) in humans. Our previous studies have shown Map to be present in U.K. rivers due to land deposition from chronic livestock infection and runoff driven by rainfall. The epidemiology of CD in Cardiff showed a significant association with the River Taff, in which Map can be detected on a regular basis. We have previously hypothesized that aerosols from the river might influence the epidemiology of CD. In this preliminary study, we detected Map by quantitative PCR in one of five aerosol samples collected above the River Taff. In addition, we examined domestic showers from different regions in the U.K. and detected Map in three out of 30 independent samples. In detecting Map in river aerosols and those from domestic showers, this is the first study to provide evidence that aerosols are an exposure route for Map to humans and may play a role in the epidemiology of CD

    Carbapenemase-producing Enterobacteriaceae in hospital wastewater:a reservoir that may be unrelated to clinical isolates [star]

    Summary Background: Carbapenemase-producing Enterobacteriaceae (CPE) are an emerging infection control problem in hospitals worldwide. Identifying carriers can help reduce potential spread and infections. Aim: To assess whether testing hospital wastewater for CPE can supplement patient based screening for infection prevention purposes in a hospital without a recognised endemic CPE problem. Methods: Wastewater collected from hospital pipework on 16 occasions during February-March 2014 was screened for CPE using chromID®CARBA agar and chromID®CPS agar with a 10 μg ertapenem disc and combination disc testing. MICs were determined using British Society for Antimicrobial Chemotherapy methodology and carbapenemase genes detected by PCR or wholegenome sequencing. Selected isolates were typed by PFGE. Findings: Suspected CPE were recovered from all 16 wastewater samples. Of 17 isolates sent to Antimicrobial Resistance and Healthcare Associated Infections Reference Unit, six (four Citrobacter freundii and two Enterobacter cloacae complex) were New Delhi metallo–beta-lactamase (NDM) producers and the remaining 11 (six Klebsiella oxytoca and five Enterobacter cloacae complex), Guiana-Extended-Spectrum-5 (GES-5) producers, the first to be described in Enterobacteriaceae in the UK. The four NDM-producing C. freundii, two NDM-producing E. cloacae complex and 4/5 GES-5-producing E. cloacae complex were each indistinguishable isolates of the same three strains, whereas the six GES-5-producing K. oxytoca overall shared 79% similarity. Conclusion: CPE are readily isolated from hospital wastewater using simple culture methods. There are either undetected carriers of CPE excreting into the wastewater, or these CPE represent colonisation of the pipework. Surveillance of hospital wastewater for CPE does not appear helpful for infection control purposes

    ‘Making the dust fly’: (A case study of) design research promoting health and sustainability in addressing household infections

    Global health crisis, such as antimicrobial resistance, threaten planetary health, as they have a direct impact on the environment, as well as to humans and animals. Personal and environmental hygiene form the best and most natural ways of reducing home infections and hence the need to take antibiotics. Despite this our understanding of cleaning in the home and interventions on home cleaning are limited. In this paper we present a project, which combined design research with environmental microbiology, to address this issue and to co-design sustainable cleaning interventions for human and planetary health. We focus on the design of a co-design workshop which led to the development of cleaning interventions tested for a month by several households. We share the challenges faced and the lessons learnt, which we envisage will help guide design researchers moving into this exciting research field of planetary and human health

    Challenges and Opportunities in Conducting and Applying Design Research beyond Global North to the Global South

    With an increase in international projects, comes increased opportunities for design researchers to contribute to research that tackles Sustainable Development Goals in the Global South. Nevertheless, the role of design within this context is disparate and detached, with very limited design literature on the subject. As such, this paper has identified the key challenges and barriers that design researchers face in conducting and applying design research in a Global South Context. Based on a workshop with design research experts doing research in Global South, the literature and lessons learnt from running a design-led project in Ghana, we are presenting and discussing 45 challenges grouped under 13 themes. We envisage that these will form the first step towards developing strategies for mitigating these challenges and help current and future design researchers to engage further in international research in the Global South

    Combining design research with microbiology to tackle drug-resistant infections in different home environments in Ghana:Challenging the boundaries of design thinking

    The aim of this paper is twofold. Firstly, to present design research work that crosses into specialist medical areas, in this instance that of antimicrobial resistance (AMR); and secondly, to contribute to the debate on the need for design driven research to explore alternative ways of delivering healthcare by focusing on preventive design responses. These points are addressed in the exploration of an international design-led research project, Dust Bunny. The project uniquely combines design research and microbiology to provide an informed assessment of societal practices in domestic cleanliness and to co-create novel solutions that reduce infections in the home. The findings and experiences from this unique transdisciplinary collaboration (design, microbiology, epidemiology and social science) reveal several insights on the challenges and opportunities on challenging the boundaries of design thinking
